Very easy Group Homework due Week TWO lab

Your task here is to get to know your textbook and lab manual so that you are able to use them as tools for the accomplishment of your projects AS WELL AS helping yourself to answer questions about lecture content.

Grading: you must correctly answer 30% of the offered points to be eligible for having your score curved to a hundred-point scale. (Curving protects you from me if I am too tough; and me from you if you are too smart! The average grade will curve to 80, so half the class will be A's and B's, and half below that.) Always beware that real life is filled with diversions, falcities and irrelevancies. Dr V's quizzes attempt to mimic life! Some questions below might have multiple answers or no answer at all. Be brave, and write "not in book" or some such thing if that is really the answer. Don't forget your texts have indexes at the back!

  1. (15 pts) What chapters in your textbook and lab manual have to do with decontamination of surfaces?

  2. (15 pts) What chapters in your textbook and lab manual have to do with self-sterilizing surfaces?

  3. You will need to make some agar-filled petri plates. ((NOTE: YOU will have to do all these procedures for real. No one can be called a microbiologist if they cannot do these fundamental procedures!)) The overall task can be divided into components. I know that most of you have never done this before. But there are many things you hadn't done before, yet you discovered directions and then followed those directions.

    1. (15 pts) Which medium to use, and how much to use per liter (or other concentration values)? You need a medium upon which a great many different bacterial species will grow. (Give-away hint:) A common one is based on beef or chicken broth and is called "nutrient broth." When the non-nutritious gelling agent agar is added, it is called nutrient agar. List procedure, book and page numbers.

    2. (15 pts) Sterilization. Which of the several methods of sterilization listed in your text and lab manual would you find easiest to use? List procedure, book and page numbers.

  4. (25 pts) (This question tests whether you can stretch your analytical skills. As a beginning beginner, can you fathom what is happening in the following procedure?) Mary Lou got diverted one day with the thought of seeing for herself just how germ-laden money really was. he placed all the current denominations of coins (taken directly from her purse) carefully onto the surface of a sterile plate containing nutrient agar (Fig, left side). In an open space, she lightly touched her finger. After allowing them to sit there for 5 minutes, she inverted the plate so that all the coins dropped off the agar onto the bench (Fig, middle). She recovered the plate and incubated it overnight at 37c. The next morning she was very depressed because she thought she had done something wrong as there was only one place where bacterial colonies were growing on the plate (Fig, right side). Jason and Wilbie saw it and got really excited. What did they observe which Mary Lou did not?

  5. (15 pts) Where in your text and lab manual is there anything about soil microbiology?
