Standardized Bacteriological Media Recipes

All weights are for a final volume of 100 mL using TAP water.
(To make solid media, add 2 gms of pure agar per 100 mL.)

L-Broth ("Luria" Broth)

0.3 gm glucose (dextrose)
0.7 gm tryptone
trace of thiamine (vit B1)*


MacConkey Agar

5 gm MacConkey Agar powder
(added directly to your autoclaving container)
(Make sure that the powder contains the sugars you want it to have!)

0.3 gm glucose
2 gm Casamino Acids
trace of thiamine (vit B1)*



* While E.coli can make a little thiamine on their own, they can not make enough for rapid grow. A speck about the size of a grain of sugar is sufficient for a liter of medium.


Nutrient Agar

(Note: this already contains agar!)
Read the bottle's label for amounts needed
use tap water for the growth minerals needed

An alternative to "NA"

"TSA" = tryptic soy agar
