Standardized Common Media Recipes

ALL WEIGHTS ARE FOR A FINAL VOLUME OF 100 MILLILITERS
(To make solid media, add 2 gms pure agar per 100 ml.)
ALWAYS USE TAP WATER FOR ITS TRACE MINERALS
(Remember that you are preparing food!)

L-Broth ("Luria" Broth)

0.3 g Glucose
0.7 g Tryptone
speck of thiamine (vitamin B1)
 

MacConkey Agar

5 g MacConkey Agar powder
(added directly to your autoclaving container)
(Make sure that the powder contains the sugars you want it to have!
If it doesn't, add 0.7 gm of that sugar.)
 

C A A

0.3 g Glucose
2 g Casamino Acids
speck of Thiamine (Vitamin B1)
 

M S (Mineral Salts)

Autoclave "A" separately; allow to cool before continuing:
A: 90 ml of tap water (trace elements)
5 ml from Bottle labelled "+/Cations"
0.05 g Na3Citrate and dissolve completely

Filter sterilize via syringe/"acrodisc":
B: 5 ml from Bottle labelled "-/Anions"
speck of Thiamine (Vitamin B1)
 

G M S

Add to MS, above:
Dissolve 0.5 g Glucose into "B"
